I've messed around with the new markers, etc, and have tried just freeform fishing, but, nothing. Am I missing something here or is fishing broken?

I'm trying to specifically mine Mythril Ore, but the bot keeps trying to run away from the location. I also cannot set a "starting" marker for the bot. And it doesn't seem to recognize the priority list.

head over to the tutorial page and check out the gathering guide. You need to set your marker mode to list and drop a marker near the nodes you want to gather.
